The Science behind Orlistat Capsules
Orlistat is a synthetic derivative of lipstatin, a natural product isolated from the bacterium Streptomyces toxytricini.
The key to its action lies in its ability to interact with enzymes called lipases—specifically, gastric and pancreatic lipases.
How Orlistat Works
The mechanism of action for Orlistat is direct and localized to the gastrointestinal (GI) tract.
- Enzyme Inhibition: When you take Orlistat with a meal containing fat, the medication travels to your stomach and small intestine. Here, it forms a covalent bond with the active site of the gastric and pancreatic lipases.
- Blocking Fat Digestion: By binding to the lipases, Orlistat inactivates these enzymes.
Normally, lipases break down large dietary fat molecules (triglycerides) into smaller, absorbable components called free fatty acids and monoglycerides. With the enzymes inhibited, this breakdown process is significantly reduced. - Preventing Absorption: Since the fat molecules remain largely undigested, they are too large to be absorbed through the wall of the small intestine and into the bloodstream.
- Excretion: As a result, approximately one-third of the fat from the food you eat is not absorbed into your body.
This unabsorbed fat simply passes through your digestive system and is eliminated in your stool.
This reduction in absorbed dietary fat creates a caloric deficit, which, when combined with a reduced-calorie, low-fat diet and exercise, leads to weight loss.
